Skyros is an ancient sovereignty that existed sometime in Equestria's distant past. It was the birthplace and foalhood home of Princess Celestia, Princess Luna, and Opaline Arcana. It's strongly implied that Skyros was a nation or city-state comprised entirely of or at least ruled by Alicorn ponies. In the nation's later years, Celestia and Luna were strongly lauded by the other subjects of the land for their unique connections to the Sun and Moon respectively. This attention largely overshadowed their young acquaintance Opaline and the Royal Sisters did not help matters by ignoring Opaline and refusing to accept her as a friend.
Celestia and Luna eventually left Skyros and head to Equestria, roughly 1,111 years before the Reign of Twilight Sparkle. Celestia and Luna, having been mentored by the Unicorn Pony Starswirl the Bearded, and having befriended a number of other Unicorns, would use their power to lead the pony kingdom of Equestria, all while Opaline eventually got banished by Skyros during Twilight's reign. She would later embark on a campaign to fracture and conquer Equestria for herself, though her ambitions would be halted for another few centuries due to the efforts of Celestia's student Twilight.
Very little evidence remains of Skyros ever having existed in Equestria. Only Opaline, Mane 6 and the Dragons know of it. Written artifacts are just as rare, with only one book in the Royal Archives in the Pegasi City of Zephyr Heights detailing the history of the Alicorns. But even that text has been diminished. It's highly likely Twilight Sparkle was aware of Skyros, but even in her time, it rarely arose in discussion.
